Clinical Relevance

This Sample Prep Set allows the quantitative determination of vanillylmandelic acid (VMA), homovanillic acid (HVA), 5-hydroxyindoleacetic acid (5-HIAA) in patient urine samples by LC-MS/MS. It is used for the diagnostics of neuroblastoma and carcinoid tumours.

Pre-analytic Treatment

24 h urine is collected in a suitable container with 10 ml of 10–25 % HCl (VMA, HVA) or 10 ml glacial acetic acid (5-HIAA).
Spontaneous urine can be also analysed (see instruction manual).

Patient specimens are stable for up to 14 days at +2 to +8 °C in amber coloured vials.
For longer storage periods keep samples frozen below -18 °C.

Sample Preparation
  • Pipette 50 μl sample/calibrator/MassCheck® control into a well of the collection plate.
  • Add 25 μl Internal Standard Mix.
  • Add 425 μl Stabilisation Buffer.
  • Seal collection plate with adhesive seal.
  • Agitate collection plate for 2 min at 1200 rpm (orbit 2 mm) and transfer to autosampler.
  • Inject eluates into the LC-MS/MS system.
Sample Stability The prepared samples are stable for up to 5 days (VMA and HVA) or up to 3 days ( 5-HIAA) at +2°C to +8°C in amber coloured vials. For longer storage periods keep samples frozen below -18°C.
