Hydrolysis efficiency monitoring for every sample

The new, patented Chromsystems hydrolysis monitoring checks the efficiency of hydrolysis in every individual patient sample and also captures sample-specific effects. This allows samples with incomplete hydrolysis to be identified.

Functional Principle

An isotope-labeled hydrolysis marker (codeine-D6-glucuronide)
is added to each sample. During incubation, this marker – like the other glucuronides in the sample – is cleaved (hydrolysed) by the hydrolysis
enzyme. A hydrolysis reference contains the same amount of hydrolysis marker but is processed without enzyme. As a result, the marker in it remains completely uncleaved.

Comparing the amount of uncleaved marker in the sample and the reference allows the hydrolysis efficiency to be determined. The less uncleaved marker remains in the sample, the more efficient the hydrolysis was.

Hydrolysis duration

Provided no inhibitory effects occur, the optimised enzyme enables complete hydrolysis in just 30 minutes – even for analytes that are difficult to hydrolyse, such as codeine. This helps to ensure reliable and complete hydrolysis.
