Clinical relevance
This assay allows the routine analysis of amino acids, succinylacetone, free carnitine, acylcarnitines, lysophospholipids, nucleosides, creatine and creatine-related metabolites in dried blood spots (DBS) for the newborn screening of metabolic disorders such as amino acidopathies, organic acidurias, fatty acid oxidation disorders, and purine or creatine metabolism disorders by MS/MS.

It is intended for screening of newborns for inherited metabolic disorders within the first few days of life.

Specimen Dried blood
Sample Preparation

The information on the sample preparation presented here is not sufficient for use in the laboratory. For a detailed step by step description, please refer to the instruction manual.

• Punch a dried blood spot disk out of the filter card into a 96 Well Filter Plate.
• Add 200 µL Internal Standard Mix, seal with a Protective Sheet and agitate for 20 min.
• Draw through the supernatant into the 96 Well Plate by centrifugation. Take off the 96 Well Filter Plate. Seal the 96 Well Plate with new Protective Sheet.
• Add 75 µL Extraction Buffer Succinylacetone, then 75 µL Internal Standard Succinylacetone to the remaining dried blood spot disk. Seal the 96 Well Filter Plate with a Protective Sheet. Agitate for 30 min at 45 °C.
• Combine both extracts and seal the 96 Well Plate.
• Agitate for 45 min at +45 °C.
• Inject 10 µL of the eluate into the MS/MS system.
