Clinical relevance

This Sample Prep Set – in combination with the chromatographic platform (order no. 80000) – allows the quantitative determination of adrenaline (epinephrine), noradrenaline (norepinephrine), dopamine, 3-methoxytyramine, metanephrine, normetanephrine and serotonin in patient urine samples by LC-MS/MS.

It is used as a test for patients in whom the urine levels of these biogenic amines are of clinical importance.

These are the analytical column, mobile phases A and B and the rinsing solution.

Lower and Upper Limit of Quantitation

Noradrenaline (norepinephrine): 4.00 µg/l – 1600 µg/l
Adrenaline (epinephrine): 2.50 µg/l – 1600 µg/l
Dopamine: 13.5 µg/l – 3000 µg/l
Normetanephrine: 3.50 µg/l – 5000 µg/l
Metanephrine: 2.00 µg/l – 5000 µg/l
3-Methoxytyramine: 3.00 µg/l – 2600 µg/l

Sample Preparation

  • Pipette 200 µl of the sample/calibrator/MassCheck® control into each Sample Clean Up Column.
  • Add 50 µl Internal Standard Mix and 600 µl Neutralisation Buffer.
  • Check for correct colour.
  • Centrifuge 5 min at 150 x g, discard effluent.
  • Add 0.9 ml Wash Buffer 1 and centrifuge 5 min at 150 x g, discard effluent.
  • Add 2 x 0.9 ml Wash Buffer 2 and centrifuge each time 1 min at 150 x g, discard effluent.
  • Centrifuge 1 min at 2000 x g to dryness, discard effluent.
  • Put Sample Clean Up Columns on new glass collection vials (e.g. glass autosampler vials).
  • Add 500 µl Elution Buffer and centrifuge 2 min at 100 x g.
  • Add 500 µl Dilution Buffer to each glass vial.
  • Seal each glass vial.
  • Shortly vortex each glass vial and transfer vials to autosampler.
  • Inject up to 10 µl.
