Clinical relevance

This assay allows the quantitative determination of free metanephrine, free normetanephrine and free 3-methoxytyramine in human plasma samples by LC-MS/MS. The assay can be employed as an aid to diagnose and monitor suspected pheochromocytoma or paraganglioma. Furthermore, the assay is an aid to diagnosing and monitoring suspected neuroblastoma.

Assay characteristics

Sample clean-up is performed by efficient solid phase extraction either in 96 SPE well plates or SPE columns (Sample Clean Up Columns). Solid phase extraction does not require equilibration or conditioning steps and accomplishes analyte enrichment without a vaporisation step. Each analyte is measured in relation to a specific isotope-labelled internal standard to ensure that reproducible and reliable quantitative results are obtained. The 6PLUS1® Multilevel Calibrator Set and MassCheck® controls are obtained from human plasma to minimise matrix-induced differences.

  • Pipette 500 µl Dilution Buffer and 25 µl Internal Standard Mix into each well of the 96 SPE Well Plate.
  • Add 500 µl of the sample/calibrator/MassCheck® control, centrifuge, discard effluent.
  • Add 0.9 ml Wash Buffer 1, centrifuge, discard effluent.
  • Add 2 x 0.9 ml Wash Buffer 2, centrifuge, discard effluent.
  • Add 250 µl Elution Buffer and centrifuge.
  • Inject 5–50 µl into the LC-MS/MS system.
