Clinical relevance

β-carotene (provitamin A) is a precursor form of vitamin A, and is an essential nutrient for the human diet. Like glutathione or vitamin C, it possesses antioxidative properties. β-carotene is a herbal dye used as a colouring in the food industry and in vitamin products.

β-carotene can only be processed by the body when absorbed together with fat. Any additional high-dosed preventive therapy should be used with caution to prevent a possible overdose. Additional use of high dose β-carotene over several years combined with nicotine and alcohol consumption may even increase the risk of lung or bowel cancer. For this reason, medications that contain β-carotene are labelled with a warning for smokers. Furthermore, drugs with high concentrations of β-carotene are no longer allowed to be prescribed to smokers either.

This reagent kit is designed for easy and reliable determination of β-carotene in serum/plasma. The sample clean up procedure combines a fast precipitation and extraction step. The resulting extract is soluble in the mobile phase, so no evaporation is necessary. The chromatographic determination is run on an isocratic HPLC system with UV/VIS detection. A special RP-chromatography ensures a reliable separation of α-, cis-β-, and all-trans-β-carotene as well as some further carotenoids in less than 10 minutes. The analytes are quantified by the inclusion of an internal standard, which is a non-naturally occurring carotenoid-derivative, so that only a single detection wavelength is required. The high stability of the processed samples allows for easier performance of large sample batches.

Sample Preparation

 

  • Pipette 100 µl serum/plasma into a light protected reaction vial.
  • Add 50 µl internal standard, mix briefly.
  • Add 50 µl Precipitation Reagent, mix briefly (vortex), do not centrifuge!
  • Add 200 µl Extraction Buffer, mix about 30 s (vortex).
  • Centrifuge 10 min at 16000 x g.
  • Transfer the supernatant into a light protected autosampler vial (glass!).
  • Inject 50 µl into the HPLC system.
