Vitamin B1 in Whole Blood and Vitamin B6 in Whole Blood/Plasma - HPLC

Order No.: 52052/Premix/RUO, for 100 tests
Parameters:
Vitamin B1 (TPP, thiamine pyrophosphate), Vitamin B6 (PLP, pyridoxal 5’-phosphate)

Each analyte safeguarded by a specific internal standard
No post-column derivatisation required
One sample preparation and one run for both analytes
Simplified sample prep with Pre-mixed Tubes

Vitamin B1 (TPP, thiamine pyrophosphate)

Vitamin B6 (PLP, pyridoxal 5’-phosphate)

This Chromsystems assay facilitates the combined analysis of vitamin B1 in whole blood and vitamin B6 in whole blood and plasma research samples. The efficient sample preparation with Pre-mixed Tubes processes different matrices, thus whole blood research samples can easily be measured in the same sequence with plasma research samples. Vitamin molecules are derivatised during sample preparation which renders the common post-column derivatisation unnecessary. The separation takes place with a binary gradient. Both analytes are safeguarded by specifically developed internal standards. Quality controls are also available separately.

Specimen Whole Blood and Whole Blood/Plasma
Sample Preparation
  • Mix 200 μl research sample/calibrator/control with 100 μl Internal Standard and 300 μl Precipitation Reagent in an amber vial (light protection) and shake 30 s (vortex).
  • Centrifuge for 5 min with at least 14000 x g.
  • Pipette 100 μl of the derivatisation mix into a labelled Pre-mixed Neutralisation Tube. Add 250 μl of the supernatant obtained above, mix briefly.
  • Incubate for 25 min at 60 °C (water bath).
  • Cool research sample for 10 min at +2 to +8°C, then centrifuge for 2 min with at least 14000 x g.
  • Transfer supernatant into a light protected autosampler vial, inject 25–50 μl into the HPLC system.
Run Time 7 - 9 min
Injection Volume 25-50 µl
Flow Rate 1.5 - 2.3 ml/min
Column Temperature ambient (~ 25 °C)
