This Chromsystems reagent kit is designed for the specific determination of the antiepileptic drugs carbamazepine, oxcarbamazepine, ethosuximide, lamotrigine, primidone, phenobarbital, phenytoin, sultiame and zonisamide in serum or plasma. In addition, the analogues carbamazepine-10,11-epoxide, 10,11-dihydro-10,11-dihydroxycarbamazepine, 10-OH-carbamazepine and 2-ethyl-2-phenylmalondiamide (PEMA) can be analysed.
Sample preparation is based on a simple but efficient precipitation step. To separate and quantify all analytes a chromatography with high resolution is used.

Specimen Serum/Plasma
Sample Preparation
  • Pipette 100 μl serum/plasma (standards, controls, research specimens) and 150 μl Internal Standard into a labelled reaction vial, mix briefly (vortex).
  • Add 50 μl Precipitation Reagent, mix 1 min (vortex).
  • Centrifuge 10–15 min at 9000 x g.
  • Mix 100 μl of the supernatant with 100 μl Stabilisation Buffer.
  • Inject 20 μl into the HPLC system.

Flow Rate High Resolution: 1.2 ml/min
Column Temperature ambient (~25 °C)

Additional Info Analysis of antiepileptic drugs requires a simple isocratic HPLC system with HPLC pump, injector and UV detector. The use of a thermostat-controlled column oven avoids temperature variations and optimises the stability of the separation.
